Identification of Endophytic Microbiota of Phytoplasma-Infected Russian Olive Trees “Elaeagnus angustifolia L.” in the Northwest of Iran

In this study, Russian olive trees exhibiting witches’-broom symptoms were collected from urban green areas in Tabriz, the northwest of Iran. PCR analysis confirmed that phytoplasma caused disease and, according to resulting Sanger sequencing electropherogram, a mixed infection with two or more species within was revealed. Next-generation analyses, using Illumina method, performed on total DNA infected plants recognize microbial genomic content and assemble whole genome causative pathogen(s). The use MetaphlAn2 Kraken2 analyze composition revealed very diverse unique compositions different Prokaryotic Eukaryotic plants. Several bacteria fungi discovered inside samples, among which Mycoplasmatota significantly dominating. Interestingly, results also high level endosymbiotic Archaea (Methanobacteria) contents samples.
